Borough travel to Braintree on 29 April and play the return leg in East Sussex on 3 May.
Eastbourne have come close to making the non-league football's top tier before, losing in the 2005 play-off final to Altrincham at Stoke's Britannia Stadium.
Wilson says they have progressed massively since then: "In our last play-off final we weren't the best prepared. This year we're more mature as a club.
"Everything about it is better. It's pointing to a victory for Eastbourne Borough."
